Background and Objectives: Tetracyclines are antibiotics that are widely used. Tetracycline, easily passes the blood-brain barrier and protects the nervous system due to its specific chemical structure. Vitamin E is one of the fat-soluble vitamins. This vitamin helps prevent the progression of Alzheimer's disease. The purpose of the present study is to investigate the effects of tetracycline and vitamin E on spatial memory, learning, and depression.   Methods: In this experimental study, 21 male Wistar rats were randomly divided into 3 groups of 7 each. The effect of tetracycline and vitamin E on memory, learning, and depression was investigated using 8-arm radial maze and elevated plus maze. Data were analyzed by one-way ANOVA and Tukey's statistical tests. Statistical significance level was considered p<0.05.   Results: After 21 days of treatment, it was shown that tetracycline antibiotic has a significant effect on memory. Also Vitamin E significantly reduced depression and its protective effect decreased the adverse effect of tetracycline. In the treatment group, vitamin E along with tetracycline had a significant effect on memory loss.   Conclusion: The results of the present study indicated that use of vitamin E can reduce depression by its antioxidant and protective effects. Tetracycline also significantly increases memory. Therefore, according to the results, it is suggested that tetracycline be used along with vitamin E to reduce negative effects of the drug.
